S-palmitoylation occurs on cysteine residues and involves a thioester linkage. In the cases where it has been investigated, S-palmitoylation is a reversible modification. Palmitoylation can affect proteins in many different ways, including regulating membrane attachment, intracellular trafficking, and membrane micro-localisation.
